ABSTRACT

This study aimed to investigate the effects of concentration, intestinal section and borneol on the intestinal absorption of salvianolic acids. The experiment not only studied the intestinal absorption properties of three concentrations of rosmarinic acid, salvianolic acid B and salvianolic acid A at duodenum, jejunum and ileum, but also of salvianolic acids compatible with borneol at different concentrations using single-pass intestinal perfusion model in rat with phenol red as the marker. The results showed that salvianolic acids was stable under weak-acid condition and affected by metabolism enzyme; The Peff and Ka significantly different among three concentrations of rosmarinic acid and salvianolic acid B, whose intestinal absorption were saturated in high concentration, suggesting that the transport mechanisms of rosmarinic acid and salvianolic acid B were similar to active transport or facilitated diffusion; However, there was inconspicuousness in the Peff and Ka of salvianolic acid A at different concentrations, whose absorption was not saturated in high concentration, indicating that the transport mechanisms of salvianolic acid A was passive diffusion; The Peff and Ka in the ileum obviously higher than those in the duodenum and jejunum, namely the ileum was the best absorption section; When concentration of borneol increased, the enhancing effect of intestinal absorption of salvianolic acids increased, but significantly decreased when borneol increased to some degree. The enhancing effect of medium borneol concentration was the optimum. This implied that borneol can enhance the intestinal absorption of salvianolic acids, and the capacity of enhancing effect was influenced by the concentration of borneol.

ABSTRACT

Objective To study the different extraction methods for the content of tanshinoneⅡA in water decocted Radix Salviae Miltiorrhiza, in order to develop comprehensive utilization of Salvia miltiorrhiza. Method Tanshinone ⅡA was extracted by supercritical carbon dioxide fluid and compared with ultrasonic extraction. Result When the extraction pressure was 35 MPa, extraction temperature was 40 ℃, extraction time was 2 h and entrainer (ethanol) was 100 %, the yields of the tanshinone ⅡA was up to 3.87 mg, while the ultrasonic extraction was 2.89 mg. Conclusion TanshinoneⅡA extracted by supercritical carbon dioxide fluid was better than ultrasonic extraction and had higher purity. The comprehensive utilization of Salvia miltiorrhiza is available.

ABSTRACT

Objective To optimize the preparation process of Xuefuzhuyu Mixture. Methods Using the extracting rate of paeoniflorin as main indicator, the content of extracting as reference indicators, the extracting process was studied by L9(34) orthogonal method. Enrichment technique was reviewed by concentrated methods and levels. Centrifugal process was reviewed by speed and time. Results The optimum extracting procedure of Xuefuzhuyu Mixture was by 10 times of water soaking for 1 h, then decocting for 60 min and 2 times. Using decompressed enrichment, mixture was enriched 1 g herbs/1 mL. Clarify solution will be received if the condition up to 4 000 r/min and run more than 7 min. Conclusion The preparation process is reasonable and viable.

Object To observe the effect of tetrandrine (Tet) on the activity of cholinesterase (ChE) inhibited by organophosphate. Methods Activity of ChE in human blood was determined with Ellman's method and used as an index. Results The cumulative dose effect curves for inhibitory effect of dipterex on ChE were shifted to right and the maximal inhibitory effect of dipterex was obviously declined by Tet. ABSTRACT

AIM Taurine can regulate Asp、 Glu、 GABA from rat cortical synaptosomes. But its mechanism is unclear. The release regulated effects were investigated through analysis of GABA receptors.METHODS Bicuculline、Phaclofen、Baclofen were added in a Krebs Ringer buffer with resuspended synapotomes.Endogenous Asp、 Glu and GABA release during the 5 min superfusion were measured by high perfusion liquid chromatography using percolumn durivatization with Dans Cl. RESULT Phaclofen,but not bicucullion baclofen, counteracted the inhibition of GABA overflow,although the inhibition of Asp and Glu overflow was not attenuated. CONCLUSION Taurine inhibits the depolarization evoked release of GABA through the activation of presynaptic autoreceptors and taurine also acts on presynaptic sits of Asp Glu nerve terminals to inhibit their evoked release in rat cerebral cotex.
